πΏ Reviewer Guide & Digital Calipers
How to inspect and verify stomata measurements to benchmark precision for ploidy sizing.
1. Anatomical Measurement Dimensions
π Major Axis (Full Stoma Length)
Spans between the two guard cell junctions (poles). Key trait for ploidy level discrimination (diploids 2x vs tetraploids 4x).
π Minor Axis (Full Stoma Width)
Cross-sectional diameter perpendicular to the major axis at the centroid. Includes both guard cells and the pore.
π΅ Central Pore Aperture Width
Inner central opening diameter measured along the minor axis. Critical proxy for stomatal conductance ($g_{\text{smax}}$).
π’ Guard Cell Width
Radial thickness of one guard cell: $\frac{L_{\text{minor}} - P_{\text{width}}}{2}$. Turgor regulation trait.
2. Keyboard Shortcuts
| 1 | High Precision (Suitable for ploidy sizing) |
| 2 | Minor Deviation (Acceptable for density) |
| 3 | Poor Measurement (Bad outline/pore missed) |
| 4 | False Stoma (Debris / non-stoma) |
| Enter | Save decision and advance to next stoma |
| C | Toggle Digital Caliper mode |
| X | Toggle CLAHE contrast filter |
| Space | Hold to peek at raw micrograph (hides overlays) |
| β / β | Navigate previous / next stoma |
